Pathogenic for Birt-Hogg-Dube syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_17135706)_(17147081_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing non-coding exon 1 and non-coding exon 2 of the FLCN gene. The 5' end of this event is unknown as it extends beyond the assayed region for this gene, and therefore may encompass additional genes. The 3' boundary is likely confined to intron 2 of the FLCN gene. For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with FLCN-related conditions. The region of the FLCN gene that includes non-coding exon(s) 1 has been determined to be clinically significant (PMID: 22382802, 21412933). Therefore, deletions that encompass that region are likely to cause disease.
