This variant lies in the LAMA2 gene (transcript NM_000426.4) at coding-DNA position 9168 through coding-DNA position 9169, duplicating 2 bases; at the protein level this means shifts the reading frame starting at serine residue 3057, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the LAMA2 protein in which other variant(s) (p.Lys3074Asnfs*5) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with LAMA2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ser3057Tyrfs*23) in the LAMA2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 66 amino acid(s) of the LAMA2 protein.
