This variant lies in the PDE6B gene (transcript NM_000283.4) at coding-DNA position 1133, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 378 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This premature translational stop signal has been observed in individual(s) with retinitis pigmentosa (PMID: 24938718, 27588261, 30998820). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (no rsID available, gnomAD 0.04%). This sequence change creates a premature translational stop signal (p.Trp378*) in the PDE6B gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PDE6B are known to be pathogenic (PMID: 8394174, 8595886, 22334370).
