This variant lies in the CEP290 gene (transcript NM_025114.4) at the canonical splice donor site of the intron immediately after coding-DNA position 297, deleting one base. Submitter rationale: This variant has not been reported in the literature in individuals affected with CEP290-related conditions. This sequence change creates a premature translational stop signal (p.Met100Trpfs*25) in the CEP290 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CEP290 are known to be pathogenic (PMID: 16909394, 17345604, 20690115). This variant is not present in population databases (gnomAD no frequency). This variant is also known as c.297+1del. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
