Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000094.4(COL7A1):c.4172del (p.Pro1391fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Pro1391Glnfs*8) in the COL7A1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in COL7A1 are known to be pathogenic (PMID: 16971478). This variant is not present in population databases (ExAC no frequency). For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with clinical features of epidermolysis bullosa dystrophica (PMID: 12485454).
