This variant lies in the BRCA2 gene (transcript NM_000059.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1910, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. For these reasons, this variant has been classified as Pathogenic. Variants at this splice site have been observed in individual(s) with Fanconi anemia (Invitae). In at least one individual the data is consistent with a variant at this splice site being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (ExAC no frequency). This sequence change affects an acceptor splice site in intron 10 of the BRCA2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584).
