Pathogenic for Torsion dystonia 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018105.3(THAP1):c.348del (p.Ile116fs), citing Invitae Variant Classification Sherloc (09022015): This premature translational stop signal has been observed in individual(s) with dystonia (PMID: 24976531). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Ile116Metfs*4) in the THAP1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 98 amino acid(s) of the THAP1 protein. This variant disrupts a region of the THAP1 protein in which other variant(s) (p.Arg169*) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
