A similar deletion of exons 1-6 ‚Äãhas been observed in individual(s) with Krabbe disease (PMID: 22704718). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant This variant is a gross deletion of the genomic region encompassing exon(s) 1-6 of the GALC gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 6 of the GALC gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in GALC are known to be pathogenic (PMID: 7437911, 9272171, 16607461).
