This variant lies in the SLC2A1 gene (transcript NM_006516.4) at coding-DNA position 1224 through coding-DNA position 1225, deleting 2 bases; at the protein level this means shifts the reading frame starting at phenylalanine residue 409, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Phe409Leufs*45) in the SLC2A1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 84 amino acid(s) of the SLC2A1 protein. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SLC2A1-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant disrupts a region of the SLC2A1 protein in which other variant(s) (p.Glu426*) have been determined to be pathogenic (Invitae).
