NC_000001.10:g.(?_235826240)_(235976381_?)del was classified as Pathogenic for Chédiak-Higashi syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 4-53 of the LYST gene. The 5' boundary is likely confined to intron 3. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. For these reasons, this variant has been classified as Pathogenic. This variant disrupts the C-terminus of the LYST protein. Other variant(s) that disrupt this region (Deletion (Exons 23-53), Deletion (Exons 15-53)) have been determined to be pathogenic (Invitae). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. This variant has not been reported in the literature in individuals with LYST-related conditions.
