Pathogenic for Pheochromocytoma/paraganglioma syndrome 3; Gastrointestinal stromal tumor — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003001.5(SDHC):c.214del (p.Arg72fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SDHC gene (transcript NM_003001.5) at coding-DNA position 214, deleting one base; at the protein level this means shifts the reading frame starting at arginine residue 72, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with SDHC-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Arg72Valfs*7) in the SDHC gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SDHC are known to be pathogenic (PMID: 19454582, 24758179).
