Pathogenic for Joubert syndrome; Meckel-Gruber syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_56295969)_(56296506_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with MKS1-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 2 of the MKS1 gene. This deletion is out-of-frame, and is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Loss-of-function variants in MKS1 are known to be pathogenic (PMID: 19466712, 24886560, 26490104).
