Pathogenic for Diffuse cerebral and cerebellar atrophy - intractable seizures - progressive microcephaly syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005051.3(QARS1):c.1978_1979del (p.Leu661fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the QARS1 gene (transcript NM_005051.3) at coding-DNA position 1978 through coding-DNA position 1979, deleting 2 bases; at the protein level this means shifts the reading frame starting at leucine residue 661, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Leu661Glyfs*27) in the QARS gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in QARS are known to be pathogenic (PMID: 24656866, 25471517). For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with QARS-related conditions.
