Pathogenic for DICER1-related tumor predisposition — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_177438.3(DICER1):c.5176C>T (p.Gln1726Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has been observed in individual(s) with DICER1-related pleuropulmonary blastoma familial tumor predisposition syndrome (PMID: 22180160). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Gln1726*) in the DICER1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DICER1 are known to be pathogenic (PMID: 19556464, 21266384).
