ClinVar contains an entry for this variant (Variation ID: 1456187). This variant is also known as c.1242-1243insCC, M415fsX496. This premature translational stop signal has been observed in individual(s) with clinical features of LAMB2-related conditions (PMID: 16898484, 17256789). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (no rsID available, gnomAD 0.003%). This sequence change creates a premature translational stop signal (p.Met415Profs*83) in the LAMB2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in LAMB2 are known to be pathogenic (PMID: 15367484).
