Pathogenic for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma; Arrhythmogenic right ventricular dysplasia 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.11:g.(?_7583161)_(7587903_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant disrupts the C-terminus of the DSP protein. Other variant(s) that disrupt this region (p.Lys2693Profs*3, p.Thr2733Serfs*14, p.Gln2765Alafs*23, p.Thr2634Lysfs*4) have been determined to be pathogenic (PMID: 11063735, 28527814, 21859740). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with DSP-related conditions. This variant results in the deletion of part of exon 24 (c.5671_*1792delinsAGAACAGTCTT) of the DSP gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
