RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is present in population databases (rs752651197, gnomAD 0.005%). Disruption of this splice site has been observed in individual(s) with hemophagocytic lymphohistiocytosis (PMID: 15466010, 27896523, 32542393). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1455802). Studies have shown that disruption of this splice site results in skipping of exon 5, and produces a non-functional protein and/or introduces a premature termination codon (PMID: 15466010). For these reasons, this variant has been classified as Pathogenic.
