Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.11:g.(?_16710965)_(41275270_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with CTNNB1-related conditions. This sequence change is a complex rearrangement involving exons 1-9 of the CTNNB1 gene. It does not change the copy number of any exons. Although the exact nature of the event is unknown, it is likely that this is a large inversion event including exons 1-9, that would either disrupt the transcription or result in a truncated protein product. Loss-of-function variants in CTNNB1 are known to be pathogenic (PMID: 23033978, 24614104, 25326669, 26350204, 28575650). For these reasons, this variant has been classified as Pathogenic.
