Submitter rationale: This sequence change replaces isoleucine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 156 of the TWIST1 protein (p.Ile156Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with craniosynostosis (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1455499). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Other variant(s) that disrupt this residue have been observed in individuals with TWIST1-related conditions (PMID: 11754069, 31754721; Invitae), which suggests that this may be a clinically significant amino acid residue.
