Pathogenic for Li-Fraumeni syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_7571752)_(7591611_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the TP53 gene has been identified. Loss-of-function variants in TP53 are known to be pathogenic (PMID: 20522432). The boundaries of this event are unknown as they extend beyond the assayed region for this gene and therefore may encompass additional genes. Similar deletions encompassing the full coding sequence of the TP53 gene have been reported in the literature in affected individuals (PMID: 20522432, 23172776, 26681312). Additionally, a study evaluating eight probands with deletion events involving the TP53 gene suggests that full-length deletions of the entire TP53 gene have a milder cancer-predisposition phenotype than partial deletions of TP53 (PMID: 21056402). For these reasons, this variant has been classified as Pathogenic.
