Pathogenic for Metachromatic leukodystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000022.10:g.(?_51065065)_(51065595_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exon 3 and part of exons 2 and 4 (c.464_808del) of the ARSA gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame. This variant has not been reported in the literature in individuals affected with ARSA-related conditions. For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the ARSA protein in which other variant(s) (p.Asp257His) have been determined to be pathogenic (PMID: 8982952, 10477432, 12445909). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
