This variant has not been reported in the literature in individuals affected with CNNM4-related conditions. This variant results in the deletion of exons 2-3 and part of exon 4 (c.1403-747_1743delinsTGTA) of the CNNM4 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in CNNM4 are known to be pathogenic (PMID: 19200525).
