Pathogenic for 3-Oxo-5 alpha-steroid delta 4-dehydrogenase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.11:g.(?_31751266)_(31758856_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Arg246 amino acid residue in SRD5A2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 1522235, 20493473, 26446026). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. A similar copy number variant has been observed in individual(s) with clinical features of SRD5A2-related conditions (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is a gross deletion of the genomic region encompassing exon(s) 2-5 of the SRD5A2 gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
