This variant lies in the COL6A2 gene (transcript NM_001849.4) at coding-DNA position 2133, where C is replaced by A; at the protein level this means converts the codon for tyrosine at residue 711 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Tyr711*) in the COL6A2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in COL6A2 are known to be pathogenic (PMID: 19884007, 20976770). This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with autosomal recessive COL6A2-related conditions (PMID: 29419890).
