This variant lies in the CPLANE1 gene (transcript NM_001384732.1) at coding-DNA position 9560, deleting one base; at the protein level this means shifts the reading frame starting at serine residue 3187, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change results in a frameshift in the CPLANE1 gene (p.Ser3133Ilefs*85). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 65 amino acid(s) of the CPLANE1 protein and extend the protein by 19 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This frameshift has been observed in individual(s) with clinical features of Joubert syndrome and related disorders (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. For these reasons, this variant has been classified as Pathogenic.
