Pathogenic for Intellectual disability, X-linked 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001111125.3(IQSEC2):c.4410dup (p.Asn1471fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a frameshift in the IQSEC2 gene (p.Asn1471Glnfs*136). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 18 amino acid(s) of the IQSEC2 protein and extend the protein by 117 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with IQSEC2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1454379). This variant results in an extension of the IQSEC2 protein. Other variant(s) that result in a similarly extended protein product (p.Ser1474Glnfs*133) have been determined to be pathogenic (PMID: 30666632). This suggests that these extensions are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
