NM_000057.4(BLM):c.2725C>T (p.Gln909Ter) was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q909* pathogenic mutation (also known as c.2725C>T), located in coding exon 13 of the BLM gene, results from a C to T substitution at nucleotide position 2725. This changes the amino acid from a glutamine to a stop codon within coding exon 13. This variant has been identified in conjunction with another BLM variant in an individual diagnosed with Bloom syndrome however clinical details were limited (German J et al. Hum Mutat, 2007 Aug;28:743-53). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
