Pathogenic for Myoclonic dystonia 11 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003919.3(SGCE):c.663-2A>T, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SGCE gene (transcript NM_003919.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 663, where A is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 5 of the SGCE gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with myoclonus–dystonia syndrome (PMID: 17853490). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1454062). Studies have shown that disruption of this splice site results in altered splicing, and produces a non-functional protein and/or introduces a premature termination codon (PMID: 17853490). For these reasons, this variant has been classified as Pathogenic.
