Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000094.4(COL7A1):c.3550+1del, citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Asn1186Metfs*3) in the COL7A1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in COL7A1 are known to be pathogenic (PMID: 16971478). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has not been reported in the literature in individuals with COL7A1-related conditions. It is also known as c.3550+1del. This variant is not present in population databases (ExAC no frequency).
