NC_000017.10:g.(?_41236869)_(41246163_?)del was classified as Pathogenic for Hereditary breast ovarian cancer syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). Similar deletions involving exons 10-11, which in the literature are also known as exons 11-12, have been observed in individuals with breast cancer (PMID: 18703817, Invitae). This variant is a gross deletion of the genomic region encompassing part of exon 10 and all of exon 11, including the exon 11-intron 11 boundary of the BRCA1 gene (c.1385_4186-2277del). This likely creates a premature translational stop signal and is expected to result in an absent or disrupted protein product.
