Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001298.3(CNGA3):c.1319G>A (p.Trp440Ter), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (no rsID available, gnomAD 0.003%). This sequence change creates a premature translational stop signal (p.Trp440*) in the CNGA3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 255 amino acid(s) of the CNGA3 protein. This premature translational stop signal has been observed in individuals with achromatopsia (PMID: 11536077, 24903488). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 1453201). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
