This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 6034, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 2012 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant has not been reported in the literature in individuals with FBN1-related conditions. This variant is not present in population databases (ExAC no frequency). For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Glu2012*) in the FBN1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FBN1 are known to be pathogenic (PMID: 17657824, 19293843).
