Pathogenic for Glycine encephalopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000170.3(GLDC):c.1581-42_1665+5del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the GLDC gene (transcript NM_000170.3) at 42 bases into the intron immediately before coding-DNA position 1581 through 5 bases into the intron immediately after coding-DNA position 1665, deleting this region. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant is a gross deletion of the genomic region encompassing exon(s) 13 of the GLDC gene. This deletion is out-of-frame, and is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Loss-of-function variants in GLDC are known to be pathogenic (PMID: 16601880). This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with GLDC-related conditions.
