Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022726.4(ELOVL4):c.435dup (p.Ile146fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 1452704). This premature translational stop signal has been observed in individual(s) with clinical features of autosomal recessive ichthyosis, spastic quadriplegia, and intellectual disability (PMID: 33652762). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ile146Tyrfs*29) in the ELOVL4 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ELOVL4 are known to be pathogenic (PMID: 24571530).
