Pathogenic for Glycine encephalopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000170.3(GLDC):c.1256C>G (p.Ser419Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser419*) in the GLDC gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in GLDC are known to be pathogenic (PMID: 16601880). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with glycine encephalopathy (PMID: 27362913, 29046206). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 1452234). For these reasons, this variant has been classified as Pathogenic.
