This variant lies in the COL1A1 gene (transcript NM_000088.4) at the canonical splice donor site of the intron immediately after coding-DNA position 588, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 7 of the COL1A1 gene. RNA analysis indicates that this variant induces altered splicing and likely results in a shortened protein product. This variant is not present in population databases (ExAC no frequency). Disruption of this splice site has been observed in individual(s) with osteogenesis imperfecta (PMID: 17078022, 28725987). This variant is also known as IVS7+1G>A. Studies have shown that disruption of this splice site results in skipping of exon 7, but is expected to preserve the integrity of the reading-frame (PMID: 25963598).
