This variant has been observed in individual(s) with clinical features of KCNQ2-related conditions (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with isoleucine at codon 229 of the KCNQ2 protein (p.Ser229Ile). The serine residue is moderately conserved and there is a large physicochemical difference between serine and isoleucine.
