NM_002294.3(LAMP2):c.1041del (p.Phe348fs) was classified as Pathogenic for Danon disease by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the LAMP2 protein in which other variant(s) (p.Gln353*) have been determined to be pathogenic (PMID: 27816333; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with LAMP2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Phe348Leufs*3) in the LAMP2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 63 amino acid(s) of the LAMP2 protein.
