NM_020549.5(CHAT):c.1162G>C (p.Val388Leu) was classified as Uncertain significance for Familial infantile myasthenia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CHAT protein function. This variant has not been reported in the literature in individuals with CHAT-related conditions. This variant is present in population databases (rs747366078, ExAC 0.002%). This sequence change replaces valine with leucine at codon 388 of the CHAT protein (p.Val388Leu). The valine residue is highly conserved and there is a small physicochemical difference between valine and leucine.
