Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 6.8e-05 in 251474 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for disease-causing variants in GHRHR, allowing no conclusion about variant significance. c.280C>T has been observed in individuals affected with Isolated growth hormone deficiency, type 4 (Cho_2013, Kale_2020, internal data). These data indicate that the variant may be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.281G>A, p.Arg94Gln), supporting the critical relevance of codon 94 to GHRHR protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
