This variant lies in the SQSTM1 gene (transcript NM_003900.5) at coding-DNA position 628, where C is replaced by T; at the protein level this means replaces proline at residue 210 with serine — a missense variant. Submitter rationale: This sequence change replaces proline with serine at codon 210 of the SQSTM1 protein (p.Pro210Ser). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SQSTM1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C25").
