Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001148.6(ANK2):c.7828G>T (p.Asp2610Tyr), citing Ambry Variant Classification Scheme 2023: The p.D2610Y variant (also known as c.7828G>T), located in coding exon 38 of the ANK2 gene, results from a G to T substitution at nucleotide position 7828. This exon is expressed solely in brain (Mohler PJ et al. Circulation. 2007;115(4):432-41). The aspartic acid at codon 2610 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
