Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002335.4(LRP5):c.4376G>A (p.Ser1459Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with asparagine at codon 1459 of the LRP5 protein (p.Ser1459Asn). The serine residue is moderately conserved and there is a small physicochemical difference between serine and asparagine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with LRP5-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt LRP5 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
