Uncertain significance for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma; Arrhythmogenic right ventricular dysplasia 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004415.4(DSP):c.6067G>T (p.Ala2023Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 6067, where G is replaced by T; at the protein level this means replaces alanine at residue 2023 with serine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with DSP-related conditions. This variant is present in population databases (rs776533737, ExAC 0.009%). This sequence change replaces alanine with serine at codon 2023 of the DSP protein (p.Ala2023Ser). The alanine residue is highly conserved and there is a moderate physicochemical difference between alanine and serine.
