NM_001103.4(ACTN2):c.964G>T (p.Asp322Tyr) was classified as Uncertain significance for Primary familial hypertrophic cardiomyopathy; Dilated cardiomyopathy 1AA by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid with tyrosine at codon 322 of the ACTN2 protein (p.Asp322Tyr). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and tyrosine. This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ACTN2 protein function. This variant has not been reported in the literature in individuals with ACTN2-related conditions.
