NM_017837.4(PIGV):c.1405C>T (p.Arg469Ter) was classified as Uncertain significance by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has been observed in individual(s) with PIGV-related conditions (PMID: 24129430). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs770220740, ExAC 0.009%). This sequence change creates a premature translational stop signal (p.Arg469*) in the PIGV gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 25 amino acid(s) of the PIGV protein.
