Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001451.3(FOXF1):c.121dup (p.Ala41fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with FOXF1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Ala41Glyfs*254) in the FOXF1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FOXF1 are known to be pathogenic (PMID: 19500772, 23505205).
