This variant lies in the BAG3 gene (transcript NM_004281.4) at coding-DNA position 600, where G is replaced by C; at the protein level this means replaces glutamine at residue 200 with histidine — a missense variant. Submitter rationale: This sequence change replaces glutamine with histidine at codon 200 of the BAG3 protein (p.Gln200His). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and histidine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
