Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000275.3(OCA2):c.2233A>G (p.Thr745Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with alanine at codon 745 of the OCA2 protein (p.Thr745Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine. This variant is present in population databases (rs150990900, ExAC 0.03%). This variant has not been reported in the literature in individuals affected with OCA2-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt OCA2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
