Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.136G>C (p.Ala46Pro), citing Ambry Variant Classification Scheme 2023: The p.A46P variant (also known as c.136G>C), located in coding exon 1 of the DNAH11 gene, results from a G to C substitution at nucleotide position 136. The alanine at codon 46 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
